To test the hypothesis that peripheral electric stimulation (PES) may suppress the reinstatement of morphine-induced conditioned place preference (CPP) in rats as well as the drug craving of detoxified heroin addicts in a frequency-dependent manner.
CPP model of the rat was constructed with two compartment automatic CPP apparatus, and the craving of the heroin addicts was assessed with a visual analogue scale (VAS).
(1) PES of low frequency could prevent the drug priming- or foot shock-induced reinstatement of morphine CPP; (2) this effect was naloxone-reversible, suggesting a possible involvement of endogenous opioid mechanisms; and (3) PES of low frequency could also accelerate the rate of natural decay of drug craving in heroin addicts after successful abstinence.
PES might serve as a therapeutic measure for the treatment of heroin addiction.
验证外周电刺激(PES)可能以频率依赖的方式抑制大鼠吗啡诱导的条件性位置偏爱(CPP)的恢复以及脱毒海洛因成瘾者的药物渴求这一假说。
使用两室自动CPP装置构建大鼠CPP模型,并用视觉模拟量表(VAS)评估海洛因成瘾者的渴求程度。
(1)低频PES可预防药物激发或足部电击诱导的吗啡CPP恢复;(2)该效应可被纳洛酮逆转,提示内源性阿片机制可能参与其中;(3)低频PES还可加速海洛因成瘾者成功戒断后药物渴求的自然衰减速度。
PES可能作为治疗海洛因成瘾的一种治疗措施。
