To evaluate the expression of CXCR4 in oral squamous cell carcinoma (OSCC) and its influence on the proliferation of OSCC cells.
The expression of CXCR4 was evaluated by immunohistochemical study in 91 surgically resected oral squamous cell carcinoma, and the relation between the staining and clinicopathological features was examined. Flow cytometry was used to detect CXCR4 expression in oral squamous cell carcinoma cell line. MTT assay was used to evaluate the SDF-1/CXCR4 influence on proliferation of tumor cells.
The expression of CXCR4 in OSCC was 62.6% and there was a significant association between the expression of CXCR4 and lymph node metastases (P < 0.05), tumor size (P < 0.01) and tumor histology grade (P < 0.01). In flow cytometry, a significant shift in fluorescent staining was seen in cells. In MTT assay, recombinant SDF-1 stimulated proliferation of OSCC cell and CXCR4 neutralization by monoclonal antibodies decreased proliferation.
The expression of CXCR4 was significantly correlated with the clinicopathological features and tumor proliferation. CXCR4 might be a valuable biomarker to predict the progression of OSCC.
评估趋化因子受体4(CXCR4)在口腔鳞状细胞癌(OSCC)中的表达及其对OSCC细胞增殖的影响。
采用免疫组织化学方法检测91例手术切除的口腔鳞状细胞癌中CXCR4的表达,并分析其与临床病理特征的关系。运用流式细胞术检测口腔鳞状细胞癌细胞系中CXCR4的表达。采用MTT法评估基质细胞衍生因子-1(SDF-1)/CXCR4对肿瘤细胞增殖的影响。
CXCR4在OSCC中的表达率为62.6%,且CXCR4的表达与淋巴结转移(P < 0.05)、肿瘤大小(P < 0.01)及肿瘤组织学分级(P < 0.01)显著相关。在流式细胞术中,细胞荧光染色出现明显变化。在MTT法中,重组SDF-1刺激OSCC细胞增殖,而单克隆抗体中和CXCR4则降低细胞增殖。
CXCR4的表达与临床病理特征及肿瘤增殖显著相关。CXCR4可能是预测OSCC进展的有价值生物标志物。
