Abstract

A high performance liquid chromatographic method for the determination of saikosaponin a, c and d in Radix Bupleuri was developed. YMC-pack A-314 ODS column (6 x 300mm) with an eluent consisting of acetone-water (58:42) at a flow rate of 1.2 ml/min was used. The saikosaponins were monitored by a differential refractometer without being converted into the corresponding diene derivatives in advance. Megestrol was used as the internal standard. The recovery rate of saikosaponins was higher than 95% and the coefficient of variation (CV) was 3.08%. The contents of saikosaponins a, c and d of 13 species of Radix Bupleuri collected in different districts of China were evaluated by this method.

建立了一种高效液相色谱法测定柴胡中柴胡皂苷a、c和d的含量。采用YMC-pack A-314 ODS柱(6×300mm),以丙酮-水(58:42)为流动相,流速为1.2ml/min。柴胡皂苷不经预先转化为相应的二烯衍生物,用示差折光检测器进行监测。以甲地孕酮为内标。柴胡皂苷的回收率高于95%,变异系数(CV)为3.08%。用该方法对采自中国不同地区的13种柴胡中柴胡皂苷a、c和d的含量进行了测定。
