To investigate the expressions of Toll-like receptor 4 (TLR4), myeloid differentiation factor 88 (MyD88) and nuclear factor κBp65 (NF-κBp65) in the renal tissues of obese mice induced by high-fat diet (HFD).
Forty-five C57BL/6 male mice (8 weeks old) were randomized into HFD group (n=39) and control group (n=6). The mice of the HFD group were fed high-fat and high-glucose diet for 20 weeks, and the others were given the normal diet instead. The body mass was measured weekly. At the end of the 20th week, one-third mice of the HFD group with the highest mass gain were classified as diet-induced obese (DIO) mice (n=13). The DIO mice and control mice were then sacrificed and tissues were collected for further use. The white adipose tissues (epididymal and perirenal fat pads) were removed and weighed. The levels of the serum creatinine (Cre), blood urea nitrogen (BUN) and lipopolysaccharide (LPS) were measured. The morphology of the renal tissues in both groups was studied by HE staining and microscopy. The levels of TLR2, TLR4, MyD88, NF-κBp65 and tumor necrosis factorα (TNF-α) mRNAs in the renal tissues were detected by the real-time quantitative PCR (qRT-PCR), and the levels of TLR4, MyD88 and NF-κBp65 proteins were determined by Western blotting.
Compared with the control group, the serum LPS level showed significantly increased in DIO mice; however, no difference was found between the two groups in the serum Cre and BUN as well as in renal morphological structure. What's more, except that TLR2 mRNA level was not significantly different between the two groups, the mRNA levels of TLR4, MyD88, NF-κBp65 and TNF-α in the renal tissues of the DIO mice were significantly higher than those in the control group, and the protein levels of TLR4, MyD88 and NF-κBp65 in the renal tissues of the DIO mice were also elevated significantly.
The levels of TLR4 and related proinflammatory signaling molecules increased in the renal tissues of obese mice induced by high-fat diet.

将45只8周龄C57BL/6雄性小鼠随机分为高脂饮食组(n=39)和对照组(n=6)。高脂饮食组小鼠给予高脂高糖饮食20周,其余小鼠给予正常饮食。每周测量小鼠体重。在第20周结束时,将高脂饮食组中体重增加最多的三分之一小鼠归类为饮食诱导肥胖(DIO)小鼠(n=13)。然后处死DIO小鼠和对照小鼠,收集组织以备进一步使用。去除白色脂肪组织(附睾和肾周脂肪垫)并称重。检测血清肌酐(Cre)、血尿素氮(BUN)和脂多糖(LPS)水平。通过HE染色和显微镜观察研究两组肾组织的形态。采用实时定量PCR(qRT-PCR)检测肾组织中TLR2、TLR4、MyD88、NF-κBp65和肿瘤坏死因子α(TNF-α)mRNA水平,采用蛋白质印迹法测定TLR4、MyD88和NF-κBp65蛋白水平。
与对照组相比,DIO小鼠血清LPS水平显著升高;然而,两组血清Cre和BUN以及肾形态结构无差异。此外,除两组TLR2 mRNA水平无显著差异外,DIO小鼠肾组织中TLR4、MyD88、NF-κBp65和TNF-α的mRNA水平显著高于对照组,DIO小鼠肾组织中TLR4、MyD88和NF-κBp65的蛋白水平也显著升高。
高脂饮食诱导的肥胖小鼠肾组织中TLR4及相关促炎信号分子水平升高。
