A Gram-reaction-negative, nonmotile, rod-shaped, aerobic bacterial strain, designated 15J10-9T5, was isolated from beach soil on Jeju Island, South Korea, and was characterized taxonomically using a polyphasic approach. Comparative 16S rRNA gene sequence analysis showed that the isolate belonged to the family Cytophagaceae in the phylum Bacteroidetes, and was most closely related to Spirosoma aerolatum PR1012k (96.1%) and Spirosoma panaciterrae DSM 21099 (95.7%). The detection of menaquinone MK-7 as the predominant respiratory quinone, a fatty acid profile with summed feature 3 (C ω7c/C ω6c), C ω5c, and iso-C as the major components, and a polar lipid profile with phosphatidylethanolamine as the major component also supported the affiliation of strain 15J10-9T5 in the genus Spirosoma. The G+C content of the genomic DNA was 47.7 mol %. The isolate could be clearly differentiated from its closest neighbors based on its phenotypic, phylogenetic, chemotaxonomic, and genotypic features. Therefore, strain 15J10-9T5 represents a novel species of the genus Spirosoma, for which the name Spirosoma gilvum sp. nov. is proposed. The type strain is 15J10-9T5 (=KCTC 52033 = JCM 31997).
从韩国济州岛的海滩土壤中分离出一株革兰氏阴性、无运动性、杆状、需氧的细菌菌株,命名为15J10 - 9T5,并采用多相分类法对其进行分类学特征分析。比较16S rRNA基因序列分析表明,该分离株属于拟杆菌门噬细胞菌科,与气螺旋体螺菌PR1012k(96.1%)和人参螺旋体螺菌DSM 21099(95.7%)关系最为密切。以甲基萘醌MK - 7作为主要呼吸醌、以总和特征3(C ω7c/C ω6c)、C ω5c和异C为主要成分的脂肪酸谱以及以磷脂酰乙醇胺为主要成分的极性脂质谱的检测结果也支持15J10 - 9T5菌株属于螺旋体属。基因组DNA的G + C含量为47.7 mol%。根据其表型、系统发育、化学分类和基因型特征,该分离株可与其最接近的亲缘种明显区分开来。因此,15J10 - 9T5菌株代表螺旋体属的一个新物种,为此提出名为金黄螺旋体螺菌(Spirosoma gilvum sp. nov.)。模式菌株为15J10 - 9T5(=KCTC 52033 = JCM 31997)。
