To investigate the expression of growth factor differentiation factor 15 (GDF15) in the nasal mucosa of patients with chronic rhinosinusitis (CRS) and its relationship with mucin 5AC(MUC5AC).Fifteen patients with CRS and nasal polyps, 15 patients with CRS without nasal polyps and 15 patients with normal nasal mucosa were enrolled in the study. Hisological changes of sinonasal mucosa were examined by hematoxylin and eosin (HE) stainding. The expression of total mucins was evaluated by periodic acid Schiff staining(PAS). And the expression of GDF15 and MUC5AC were measured by immunohistochemistry (IHC), and reverse transcription polymerase chain reaction (RT-PCR).The tissuse samples demonstrated mucaosal thicking, goblet cell hyperplasia, glandular hyperplasia and inflammatory cell infiltration in CRSwNP and CRSsNP. IHC revealed that GDF15 and MUC5AC expression higher in CRSwNP and CRSsNP than in normal sinus mucosa (< 0.05). qRT-PCR also displays that the relative expression levles of mRNA exprssion of GDF15 and MUC5AC were higher in CRSwNP and CRSsNP than in controls.And the expression level of GDF15 was positively correlated with that of MUC5AC mRNA in CRS.GDF15 and MUC5AC were upregulated in CRS with or without nasal polyps, indicating that GDF15 is an important factor in the process of hypersecretion of MU5AC in CRS.
探讨生长分化因子15(GDF15)在慢性鼻-鼻窦炎(CRS)患者鼻黏膜中的表达及其与黏蛋白5AC(MUC5AC)的关系。本研究纳入15例CRS伴鼻息肉患者、15例CRS不伴鼻息肉患者及15例正常鼻黏膜患者。采用苏木精-伊红(HE)染色观察鼻窦黏膜的组织学变化。用高碘酸希夫染色(PAS)评估总黏蛋白的表达。通过免疫组织化学(IHC)和逆转录聚合酶链反应(RT-PCR)检测GDF15和MUC5AC的表达。组织样本显示,CRSwNP和CRSsNP患者存在黏膜增厚、杯状细胞增生、腺体增生及炎性细胞浸润。免疫组化显示,CRSwNP和CRSsNP中GDF15和MUC5AC的表达高于正常鼻窦黏膜(<0.05)。qRT-PCR也显示,CRSwNP和CRSsNP中GDF15和MUC5AC mRNA的相对表达水平高于对照组。在CRS中,GDF15的表达水平与MUC5AC mRNA的表达水平呈正相关。GDF15和MUC5AC在伴或不伴鼻息肉的CRS中均上调,表明GDF15是CRS中MU5AC分泌过多过程中的一个重要因素。
