Misuse of pregabalin and its forensic relevance is steadily increasing. The aim of this study was to evaluate the usability of the commercially available ARKTM Pregabalin II Assay (ARK Diagnostics) for serum analysis of forensic samples. Overall, 156 samples were tested by both the immunoassay and a validated liquid chromatographic-tandem mass spectrometric (LC-MS/MS) method. Sensitivity was 100%, and specificity was 98.7% (n = 79 positive cases confirmed by LC-MS/MS in a range of 380-37,000 ng/mL). A good correlation (R = 0.73) could also be shown between quantitative immunoassay and LC-MS/MS results. In conclusion, the assay shows excellent reliability for screening of forensic serum samples.

摘要
目的：评价受试制剂苯磺酸氨氯地平片（5mg）与参比制剂苯磺酸氨氯地平片（5mg）在中国健康受试者中的生物等效性。方法：采用随机、开放、两周期交叉试验设计，20 名健康受试者随机交叉单剂量口服受试制剂或参比制剂 5mg。于给药前及给药后 0.25、0.5、0.75、1.0、1.5、2.0、2.5、3.0、3.5、4.0、5.0、6.0、8.0、10.0、12.0、16.0、20.0、24.0 和 30.0h 采集肘静脉血，测定血药浓度，计算药代动力学参数。结果：受试制剂和参比制剂的主要药代动力学参数 Cmax 分别为（5.626±0.876）和（5.633±0.910）ng/ml，tmax 分别为（1.05±0.25）和（1.00±0.23）h，AUC0-t 分别为（28.227±3.758）和（28.182±3.711）ng·h/ml，AUC0-∞ 分别为（29.102±3.850）和（29.045±3.760）ng·h/ml，t1/2 分别为（35.53±4.44）和（35.39±4.34）h。受试制剂和参比制剂的 Cmax、AUC0-t、AUC0-∞经对数转换后，其几何均值比的 90%置信区间均在 80.00%～125.00%范围内，tmax 几何均值比的 90%置信区间在 75.00%～133.33%范围内，表明受试制剂与参比制剂具有生物等效性。结论：本研究结果表明，受试制剂与参比制剂在健康受试者体内具有生物等效性。
