Elastin-like peptides (ELPs) are synthetic, repetitive sequences known for their temperature-dependent reversible self-assembly, making them environmentally friendly, biocompatible, and thermoresponsive. Despite various strategies for ELP production, the development of effective ELP supply methods remains a challenge. This chapter outlines a practical approach to synthesizing thermoresponsive ELPs using liquid-phase peptide synthesis (LPPS) with a hydrophobic benzyl alcohol support (HBA-Tag) and fragment condensation. This method effectively produces short ELPs that exhibit temperature-responsive self-assembly.
