To explore an anti-tumor effect of indomethacin on human colon adenocarcinoma cell line HCT116, and its mechanism.
MTT, agarose gel electrophoresis, TdT and flow cytometry were used to study cell proliferation and apoptosis.
Indomethacin significantly inhibited the proliferation of HCT116 cells and induced apoptosis with a "dose-time" dependent pattern. A synergic effect of inhibiting cancer cells proliferation was observed when combined with 5-Fu.
Inhibition of proliferation and induction of apoptosis contribute to the mechanisms for anti-tumor activity of indomethacin.
探讨吲哚美辛对人结肠腺癌细胞系HCT116的抗肿瘤作用及其机制。
采用MTT法、琼脂糖凝胶电泳法、TdT法和流式细胞术研究细胞增殖和凋亡情况。
吲哚美辛显著抑制HCT116细胞的增殖,并呈“剂量-时间”依赖性诱导其凋亡。与5-氟尿嘧啶联合使用时,观察到对癌细胞增殖有协同抑制作用。
抑制增殖和诱导凋亡是吲哚美辛抗肿瘤活性的机制。
