Abstract

OBJECTIVE

To study the effect of interleukin-6(IL-6) on the growth of a human lung adenocarcinoma cell line PAa in vitro.

METHODS

IL-6 receptor (IL-6R) mRNA level was detected by means of reverse transcription polymerase chain reaction (RT-PCR) and IL-6 mRNA was analyzed using Northern blotting hybridization. The production of IL-6 was detected by bioassay. The biological effect of IL-6 released by PAa cells was assessed with anti-IL-6 neutralizing antibodies.

RESULTS

PAa cells expressed IL-6R mRNA. Recombinant human IL-6 stimulated the proliferation of PAa cells. PAa cells also expressed IL-6 mRNA and produced bioactive IL-6. Treatment of PAa cells with anti-IL-6 antibodies resulted in decreased growth of PAa cells.

CONCLUSION

These results demonstrate that IL-6 acts as an autocrine growth stimulator for PAa cells in vitro.

研究白细胞介素-6(IL-6)对人肺腺癌细胞系PAa体外生长的影响。

方法

采用逆转录聚合酶链反应(RT-PCR)检测IL-6受体(IL-6R)mRNA水平,用Northern印迹杂交分析IL-6 mRNA。通过生物测定法检测IL-6的产生。用抗IL-6中和抗体评估PAa细胞释放的IL-6的生物学效应。

结果

PAa细胞表达IL-6R mRNA。重组人IL-6刺激PAa细胞增殖。PAa细胞也表达IL-6 mRNA并产生生物活性IL-6。用抗IL-6抗体处理PAa细胞导致PAa细胞生长减少。

结论

这些结果表明IL-6在体外作为PAa细胞的自分泌生长刺激因子发挥作用。
