Abstract

OBJECTIVE

To investigate the effects of hematopoietic growth factors (HGFs) on apoptosis of hematopoietic cells in myelodysplastic syndromes (MDS) patients.

METHODS

CD34+ cells in bone marrow from 12 MDS patients were purified by an immunomagnetic beads sorting system. Apoptosis of hematopoietic precursors was assayed by propidium iodine staining and flow cytometric analysis.

RESULTS AND CONCLUSION

High dose rhEpo partly suppressed apoptosis of hematopoietic cells in MDS patients. At the 7th, 10th, 14th, cultured day, the apoptotic percentages of the high dose Epo group (rhEpo 200 U/ml or 100 U/ml) were substantially lower than that of the low dose group (20 U/ml, 2 U/ml and 0.2 U/ml) and showed a dose-dependent effect with rhEpo. rhGM-CSF and rhIL-3 could also suppress hematopoietic cell apoptosis in vitro, the differences between the high dose group (rhIL-3 200 U/ml or rhGM-CSF 500 U/ml) and the low dose group (rhIL-3 50 U/ml or rhGM-CSF 50 U/ml) were significant. Extensive apoptosis of myelodysplastic hematopoietic cells were markely suppressed when rhEpo and rhIL-3 or rhEpo and rhGM-CSF were given together.

探讨造血生长因子(HGFs)对骨髓增生异常综合征(MDS)患者造血细胞凋亡的影响。

方法

采用免疫磁珠分选系统纯化12例MDS患者骨髓中的CD34+细胞。通过碘化丙啶染色和流式细胞术分析检测造血前体细胞的凋亡情况。

结果与结论

高剂量重组人促红细胞生成素(rhEpo)部分抑制了MDS患者造血细胞的凋亡。在培养的第7天、第10天、第14天,高剂量Epo组(rhEpo 200 U/ml或100 U/ml)的凋亡百分比显著低于低剂量组(20 U/ml、2 U/ml和0.2 U/ml),且呈现出rhEpo剂量依赖性效应。重组人粒细胞-巨噬细胞集落刺激因子(rhGM-CSF)和重组人白细胞介素-3(rhIL-3)在体外也能抑制造血细胞凋亡,高剂量组(rhIL-3 200 U/ml或rhGM-CSF 500 U/ml)与低剂量组(rhIL-3 50 U/ml或rhGM-CSF 50 U/ml)之间差异显著。当rhEpo与rhIL-3或rhEpo与rhGM-CSF联合使用时,骨髓增生异常造血细胞的广泛凋亡受到明显抑制。
