To investigate the vertical transmission rate of Chlamydia trachomatis (CT) in Chongqing, China.
Specimens taken from 278 women and from their 79 infants were examined by cell culture, polymerase chain reaction (PCR) and DNA sequence analysis. Chlamydia trachomatis was isolated in McCoy cell culture. CT DNA was extracted with a modified Nal method. After cloning, recombinant plasmids were used for sequence analysis with the dideoxy chain termination method.
10.8% (30/278) of the cervical cultures of pregnant women were positive for Chlamydia trachomatis, while the positive rate tested by PCR was 14.0% (39/278). The vertical transmission rate of Chlamydia trachomatis was 55.0% (11/20). The incidences of conjunctivitis and pneumonia in infants with Chlamydia trachomatis positive mothers were 27.3% and 18.2%, respectively. DNA sequences of Chlamydia trachomatis isolated from the cervix of a mother and the nasopharynx of her baby were identical.
Chlamydia trachomatis infection is quite common in Chongqing, China. Our report is the first report of CT vertical transmission proved by DNA sequence analysis.
调查中国重庆地区沙眼衣原体(CT)的垂直传播率。
对278名女性及其79名婴儿的标本进行细胞培养、聚合酶链反应(PCR)和DNA序列分析。沙眼衣原体在McCoy细胞培养中分离。采用改良的Nal法提取CT DNA。克隆后,用双脱氧链终止法对重组质粒进行序列分析。
孕妇宫颈培养物中沙眼衣原体阳性率为10.8%(30/278),而PCR检测阳性率为14.0%(39/278)。沙眼衣原体的垂直传播率为55.0%(11/20)。沙眼衣原体阳性母亲的婴儿中结膜炎和肺炎的发生率分别为27.3%和18.2%。从母亲宫颈和婴儿鼻咽分离的沙眼衣原体DNA序列相同。
沙眼衣原体感染在中国重庆地区相当普遍。我们的报告是首次通过DNA序列分析证实CT垂直传播的报告。
