To explore the expression and clinical prognostic significance of matrix metalloproteinase-2 mRNA (MMP-2 mRNA), tissue inhibitor of matrix metalloproteinase-2 mRNA (TIMP-2 mRNA), matrix metalloproteinase-2 protein (MMP-2), matrix metalloproteinase-9 protein (MMP-9), tissue inhibitor of matrix metalloproteinase-1 protein (TIMP-1), and tissue inhibitor of matrix metalloproteinase-2 protein (TIMP-2) in the hepatocellular carcinomas (HCCs).
Fifty-six specimens of HCCs from 56 patients, who were followed-up, were investigated by in situ hybridization with specific probes for MMP-2, TIMP-2, and by immunohistochemistry with anti-MMP-2, MMP-9 and anti-TIMP-1, TIMP-2 monoclonal antibody. We analyzed the data with chi-square test, spearmans correlation analysis, monovariate Kaplan-Meier plot and multivariate Cox regression analysis.
The over-expression of MMP-2mRNA, MMP-2 protein and MMP-9 protein and the imbalance between MMP-2 and TIMP-2 play pivotal roles in the degradation of excellular matrix of HCCs. MMP-2 and MMP-9 immunoreactive protein have been closely related to a shortened survival independent of major prognostic indicators in the primary HCC and increase the risk of the patients after the operation.
探讨基质金属蛋白酶-2信使核糖核酸(MMP-2 mRNA)、基质金属蛋白酶组织抑制因子-2信使核糖核酸(TIMP-2 mRNA)、基质金属蛋白酶-2蛋白(MMP-2)、基质金属蛋白酶-9蛋白(MMP-9)、基质金属蛋白酶组织抑制因子-1蛋白(TIMP-1)及基质金属蛋白酶组织抑制因子-2蛋白(TIMP-2)在肝细胞癌(HCC)中的表达及临床预后意义。
对56例接受随访的HCC患者的56份标本,采用MMP-2、TIMP-2特异性探针进行原位杂交,并用抗MMP-2、MMP-9及抗TIMP-1、TIMP-2单克隆抗体进行免疫组织化学检测。采用卡方检验、Spearman相关分析、单因素Kaplan-Meier曲线分析及多因素Cox回归分析对数据进行分析。
MMP-2mRNA、MMP-2蛋白及MMP-9蛋白的过表达以及MMP-2与TIMP-2之间的失衡在HCC细胞外基质降解中起关键作用。MMP-2及MMP-9免疫反应蛋白与原发性HCC患者生存期缩短密切相关,且独立于主要预后指标,增加了患者术后风险。
