Abstract

OBJECTIVE

To set up HSP65 PCR-RELP capillary electrophoresis method to rapidly identify the mycobacterium to species.

METHODS

A 440 base pair long part of HSP65 gene of each mycobacterium was amplified by PCR. After the amplification products were fully digested by restriction endonucleases BstE II and Hae III respectively, capillary electrophoresis with laser induced fluorescence detector was used to separate and detect the digested fragments.

RESULTS

All of the 7 tested mycobacterium species have their own unique patterns of digested fragments, all fragments can be well separated.

CONCLUSION

Capillary electrophoresis can give more higher accuracy and sensitivity in identifying mycobacterium species.

摘要

目的

建立HSP65 PCR-RELP毛细管电泳方法以快速鉴定分枝杆菌种类。

方法

通过PCR扩增各分枝杆菌HSP65基因440碱基对长的片段。扩增产物分别用限制性内切酶BstE II和Hae III充分消化后,采用激光诱导荧光检测器的毛细管电泳分离和检测消化后的片段。

结果

7种受试分枝杆菌均有其独特的消化片段模式,所有片段均可良好分离。

结论

毛细管电泳在分枝杆菌种类鉴定中可提供更高的准确性和灵敏度。
