Abstract

The Wnt family of secreted signaling molecules is essential in embryo development and tumor formation. In addition to the Frizzled family of serpentine receptors, LRP5, a member of the LDL receptor family, was recently shown to function as Wnt receptor. The finding of unique mutations in the human LRP5 gene as a modulator of bone mass establishes the importance of this novel and unexpected role of Wnt/LRP5 signaling in bone development and bone metabolism.

分泌型信号分子Wnt家族在胚胎发育和肿瘤形成过程中至关重要。除了卷曲蛋白(Frizzled)家族的蛇形受体外,低密度脂蛋白受体(LDL)家族成员LRP5最近被证明可作为Wnt受体发挥作用。人类LRP5基因中作为骨量调节因子的独特突变的发现,确立了Wnt/LRP5信号在骨骼发育和骨代谢中这一新颖且意想不到的作用的重要性。
