To identify the nucleocapsid protein of Hantaan virus (HTNV-NP)-specific T lymphocyte epitopes and analyze the epitope-specific T cell response during hemorrhagic fever with renal syndrome (HFRS).
T lymphocyte epitopes and frequencies of epitope-specific T cells were determined by ELISPOT using PBMCs from HFRS patients stimulated by individual or mixture of overlapping 15-mer peptides spanning the amino acid sequence of HTNV-NP.
Out of 10 peptide mixtures, 8 elicited strong HTNV-NP-specific responses in 18 of 47 HFRS patients, and the T cell response was found at early stage of HFRS. Moreover, 17 HTNV-NP-specific T lymphocyte epitopes were identified in 11 patients, and most epitopes were clustered near the center of NP in linear structure. Among them, 14 T lymphocyte epitopes were described for the first time.
HTNV-NP-specific T cell response can be elicited at early stage of HFRS and T lymphocyte epitopes mainly located in the center of NP, suggesting that it may play an important role in immune protection during HTNV infection.

使用来自HFRS患者的外周血单个核细胞(PBMC),通过ELISPOT法,用跨越HTNV-NP氨基酸序列的单个或重叠15肽混合物刺激,测定T淋巴细胞表位和表位特异性T细胞频率。
在10种肽混合物中,8种在47例HFRS患者中的18例中引发了强烈的HTNV-NP特异性反应,并且在HFRS早期发现了T细胞反应。此外,在11例患者中鉴定出17个HTNV-NP特异性T淋巴细胞表位,并且大多数表位在线性结构中聚集在NP的中心附近。其中,14个T淋巴细胞表位是首次被描述。
HFRS早期可引发HTNV-NP特异性T细胞反应,且T淋巴细胞表位主要位于NP的中心,提示其可能在HTNV感染期间的免疫保护中发挥重要作用。
