Abstract

PURPOSE

To investigate the effects of citicoline on upregulated clusterin and retinal damage induced by kainic acid (KA).

METHODS

KA was injected into the vitreous of rats. Effects of systemic citicoline treatments were estimated by measuring the thickness of the various retinal layers, immunoblotting, and immunohistochemical techniques.

RESULTS

One day after KA injection, the immunoreactivity of clusterin increased significantly. In rats treated with KA plus citicoline, clusterin immunoreactivity was markedly reduced compared to KA-treated rats. Western blot analysis showed that clusterin protein levels were increased in KA-treated rats, but decreased in KA plus citicoline-treated rats. Apoptotic cell death was determined by TUNEL method. Citicoline reduced the expression of clusterin, as well as the expression of TUNEL after KA injection in the rat retina.

CONCLUSION

The increased expression of clusterin following KA injection in the rat retina suggests the presence of neurodegenerative events; citicoline may provide neuroprotection against neuronal cell damage.

方法

将KA注入大鼠玻璃体。通过测量视网膜各层厚度、免疫印迹和免疫组化技术评估全身应用胞磷胆碱治疗的效果。

结果

KA注射后1天,簇集素的免疫反应性显著增加。与KA处理的大鼠相比,KA加胞磷胆碱处理的大鼠中,簇集素免疫反应性明显降低。蛋白质印迹分析表明,KA处理的大鼠中簇集素蛋白水平升高,但KA加胞磷胆碱处理的大鼠中降低。采用TUNEL法测定凋亡细胞死亡情况。胞磷胆碱降低了大鼠视网膜KA注射后簇集素的表达以及TUNEL的表达。

结论

大鼠视网膜KA注射后簇集素表达增加提示存在神经退行性变事件;胞磷胆碱可能对神经元细胞损伤提供神经保护作用。
