To study the effects of Pingchuan Recipe, a compound traditional Chinese herbal medicine for treating bronchial asthma, on macrophage inflammatory protein-1alpha (MIP-1alpha) and immunoglobulin E (IgE) contents and CD86 expression in a mouse model of bronchial asthma, and to investigate the mechanism of Pingchuan Recipe in regulating airway remodeling in mice with bronchial asthma.
Fifty BALB/c mice were randomly divided into normal control group, untreated group, dexamethasone group, low-dose Pingchuan Recipe group and normal-dose Pingchuan Recipe group. Bronchial asthma in mice was induced by intra-abdominal injection of mixed ovalbumin and stimulation by inhaling 5% ovalbumin. Mice in the normal control group and the untreated group did not receive treatment, but mice in the other groups were administered intragastrically with 50 g/ (kg * d) Pingchuan Recipe, 25 g/ (kg * d) Pingchuan Recipe and 0.75 mg/ (kg * d) dexamethasone, respectively. After consecutive treatment for 8 weeks, the MIP-1alpha contents in peripheral blood and bronchoalveolar lavage fluid (BALF) were measured by an avidin biotin complex-enzyme-linked immunosorbent assay (ABC-ELISA) system, and the IgE contents in serum and BALF were measured by ELISA method. Direct immunofluorescence-flow cytometry was used to detect the percentages of CD86(+) and CD3(-) CD86(+) cells in peripheral blood and BALF. General histological changes of the lung tissue were observed by HE staining and collagen fiber staining 12 weeks after activation.
Compared with the normal control group, the contents of MIP-1alpha, the percentages of CD86(+) cells and CD3(-) CD86(+) cells in peripheral blood and BALF, and the contents of IgE in serum and BALF were raised significantly (P < 0.01). The contents of MIP-1alpha, the percentages of CD86(+) cells and CD3(-) CD86(+) cells in peripheral blood and BALF, and the contents of IgE in serum and BALF in the treated groups were lower than those in the untreated group (P < 0.01). The contents of MIP-1alpha in peripheral blood and BALF and the contents of IgE in serum and BALF had no significant differences among the treated groups (P > 0.05). The percentage of CD3(-) CD86(+) cells in peripheral blood in low-dose Pingchuan Recipe group was lower than that in the dexamethasone group. Compared with the untreated group, the collagen deposition in the wall of bronchiole in the treated groups was decreased.
Pingchuan Recipe can decrease the contents of MIP-1alpha, IgE and the expressions of CD86 in peripheral blood and BALF, which may be one of its mechanisms in improving chronic airway inflammation and inhibiting airway remodeling.
研究治疗支气管哮喘的复方中药平喘方对支气管哮喘小鼠模型中巨噬细胞炎性蛋白-1α(MIP-1α)、免疫球蛋白E(IgE)含量及CD86表达的影响,探讨平喘方调节支气管哮喘小鼠气道重塑的机制。
将50只BALB/c小鼠随机分为正常对照组、未治疗组、地塞米松组、低剂量平喘方组和正常剂量平喘方组。通过腹腔注射混合卵清蛋白并吸入5%卵清蛋白进行刺激诱导小鼠支气管哮喘。正常对照组和未治疗组小鼠不接受治疗,而其他组小鼠分别灌胃给予50 g/(kg·d)平喘方、25 g/(kg·d)平喘方和0.75 mg/(kg·d)地塞米松。连续治疗8周后,采用抗生物素蛋白-生物素复合物-酶联免疫吸附测定(ABC-ELISA)系统检测外周血和支气管肺泡灌洗液(BALF)中MIP-1α含量,采用ELISA法检测血清和BALF中IgE含量。采用直接免疫荧光-流式细胞术检测外周血和BALF中CD86(+)及CD3(-)CD86(+)细胞的百分比。激活12周后,通过HE染色和胶原纤维染色观察肺组织的一般组织学变化。
与正常对照组相比,外周血和BALF中MIP-1α含量、CD86(+)细胞及CD3(-)CD86(+)细胞百分比,以及血清和BALF中IgE含量均显著升高(P<0.01)。治疗组外周血和BALF中MIP-1α含量、CD86(+)细胞及CD3(-)CD86(+)细胞百分比,以及血清和BALF中IgE含量均低于未治疗组(P<0.01)。各治疗组外周血和BALF中MIP-1α含量以及血清和BALF中IgE含量差异无统计学意义(P>0.05)。低剂量平喘方组外周血中CD3(-)CD86(+)细胞百分比低于地塞米松组。与未治疗组相比,治疗组细支气管壁胶原沉积减少。
平喘方可降低外周血和BALF中MIP-1α、IgE含量及CD86表达,这可能是其改善慢性气道炎症和抑制气道重塑的机制之一。
