To investigate the expression profile of adipose differentiation-related protein(ADRP)in macrophages and foam cells.
THP-1 monocytes were differentiated into macrophages in vitro, and then induced into foam cells by culturing with oxidized low density lipoproteins(ox-LDLs). Real time RT-PCR and Western blot were performed to detect the expression of ADRP gene and protein.
The expression of ADRP both mRNA and protein were significantly increased when macrophages were further induced into foam cells by ox-LDLs, however, the same results were not observed during phorbol 12-myristate-13-acetate(PMA)-induced monocyte differentiation into macrophages.
Our findings revealed an upregulation of ADRP during macrophages differentiation into foam cells.

将THP-1单核细胞在体外分化为巨噬细胞,然后用氧化低密度脂蛋白(ox-LDLs)培养诱导其成为泡沫细胞。采用实时逆转录聚合酶链反应(Real time RT-PCR)和蛋白质免疫印迹法(Western blot)检测ADRP基因和蛋白的表达。
当巨噬细胞被ox-LDLs进一步诱导成为泡沫细胞时,ADRP的mRNA和蛋白表达均显著增加,然而,在佛波酯(PMA)诱导单核细胞分化为巨噬细胞的过程中未观察到相同结果。
我们的研究结果显示在巨噬细胞分化为泡沫细胞的过程中ADRP表达上调。
