To study the expression of paxillin in the two subgroups of human breast cancer cells with high and low metastatic potentialities and the effect of paxillin on tumor metastasis and adhesion.
Human breast cancer MDA-MB-435s cells were cultured in Transwell chamber with artificial matrigel, two subgroups of MDA-MB-435s cells with different metastatic potentiality were obtained by the ability of cells penetrating artificial matrigel. The expressions of paxillin mRNA and protein were examined by Immune histochemistry, Western blot and RT-PCR. The adhesion rates of the cells were examined by MTT.
It was revealed by Immune histochemistry, Western blot, and RT-PCR that the expressions of paxillin at mRNA and protein levels were significantly higher in the cells with high metastatic potentiality. The difference was significant (t = 25.02,10.10, 17.18, P < 0.01). The adhesion rate was higher in the cells with high metastatic potentiality, and the difference was significant (F = 41.87, P < 0.01).
The expression of paxillin may play an important role in human breast cancer metastasis and adhesion.
研究桩蛋白(paxillin)在人乳腺癌高转移潜能和低转移潜能两个亚组细胞中的表达情况,以及桩蛋白对肿瘤转移和黏附的影响。
将人乳腺癌MDA-MB-435s细胞接种于铺有基质胶的Transwell小室中培养,根据细胞穿透基质胶的能力获得具有不同转移潜能的MDA-MB-435s细胞两个亚组。采用免疫组织化学、蛋白质免疫印迹法(Western blot)及逆转录-聚合酶链反应(RT-PCR)检测桩蛋白mRNA和蛋白的表达。采用MTT法检测细胞黏附率。
免疫组织化学、Western blot及RT-PCR结果显示,高转移潜能细胞中桩蛋白的mRNA和蛋白表达水平显著高于低转移潜能细胞。差异有统计学意义(t = 25.02,10.10, 17.18, P < 0.01)。高转移潜能细胞的黏附率更高,差异有统计学意义(F = 41.87, P < 0. 01)。
桩蛋白的表达可能在人乳腺癌转移和黏附中起重要作用。
