To develop a LC-MS method to determine paeoniflorin concentration in rats plasma. The method was applied to investigate pharmacokinetics of paeoniflorin in rats in vivo.
Blood samples were collected at different time after oral administration of Radix Paeoniae Alba extract at doses of 0.2, 0.4, 0.8 g x kg(-1). The paeoniflorin concentration in plasma was determined by LC-MS method. Pharmacokinetic parameters were fitted by WinNonlin 5.1 software package.
The linear range and the average recovery of paeoniflorin were 2.5-500 microg x L(-1) (r = 999 4) and more than 80% (n = 5) , respectively. The inner- and inter-days precision were both less than 15%. The T1/2 was similar. The relationship between dose and AUC showed good linearity.
The method described in this report has high sensitivity and selectivity, and was suitable for pharmacokinetic study of paeoniflorin. The kinetic process of paeoniflorin in palsma showed two-compartment model after oral administration of Radix Paeoniae Alba extract at doses of 0.2, 0.4, 0.8 g x kg(-1) to rats.
建立一种液相色谱-质谱联用(LC-MS)方法测定大鼠血浆中芍药苷浓度,并将该方法应用于研究芍药苷在大鼠体内的药代动力学。
以0.2、0.4、0.8 g·kg⁻¹的剂量给大鼠口服白芍提取物,于不同时间点采集血样,采用LC-MS法测定血浆中芍药苷浓度,用药动学软件WinNonlin 5.1拟合药代动力学参数。
芍药苷的线性范围为2.5~500 μg·L⁻¹(r = 0.9994),平均回收率大于80%(n = 5),日内和日间精密度均小于15%。半衰期相似,剂量与药时曲线下面积(AUC)呈良好线性关系。
本研究建立的方法灵敏度高、选择性好,适用于芍药苷的药代动力学研究。以0.2、0.4、0.8 g·kg⁻¹的剂量给大鼠口服白芍提取物后,芍药苷在血浆中的动力学过程呈二室模型。
