探讨聚 ADP 核糖聚合酶抑制剂 PJ34 对谷氨酸神经毒性的保护作用,为 PJ34 在缺血性脑血管病中的临床应用提供参考。
将培养的 HT-22 细胞用 5 mmol/L 谷氨酸诱导处理后,分为 PJ34 组,随后给予 50 μmol/L PARP 抑制剂 PJ34 进行保护作用处理。观察各组细胞的抑制率及 PARP 活性变化。
PJ34 组作用 24 h 后,HT-22 细胞抑制率明显降低(P<0.05)。PJ34 组 PARP 活性较对照组明显降低(P<0.05)。
PJ34 通过降低 PARP 活性增加谷氨酸诱导的 HT-22 细胞活力,从而对细胞起到保护作用。
