The present study was carried out to isolate and identify trans-tiliroside as principal compound with anti-hyperglycemic, anti-hyperlipidemic and antioxidant effects from Potentilla chinesis.
A bioactive compound, trans-tiliroside was isolated from the ethanol extract of Potentilla chinesis and its administration dose was optimized and patented. The normal, alloxan-induced diabetic mice and streptozotocin-induced diabetic rats were used to evaluate the anti-hyperglycemic, anti-hyperlipidemic and antioxidant effects of trans-tiliroside from Potentilla chinesis. Biochemical parameters were assayed in blood samples of different groups of alloxan-induced diabetic mice and streptozotocin-induced diabetic rats.
The level of fasting serum glucose levels, triglycerides (TG) and total cholesterol (TC) in alloxan-induced diabetic mice were significantly decrease after daily oral administration of trans-tiliroside in normal and diabetic mice at doses of 0.4, 0.8 and 1.6 mg/kg/day, for 15 days. Blood glucose level was significant decrease in STZ induced diabetic rats by trans-tiliroside (1.2 and 0.3mg/kg body weight for 10 weeks). The content of TC, low density lipoprotein (LDL-C) and TG levels were decreased and high density lipoprotein (HDL-C) content was increased, so lipid metabolism was improved. Moreover, trans-tiliroside revealed antioxidant activity as shown by increased activities of superoxide dismutase (SOD), decreased level of malondialdehyde (MDA) in diabetic rats. Histological morphology examination showed that the trans-tiliroside restored the damage of pancreas tissues in rats with diabetes mellitus.
Trans-tiliroside, a constituent from Potentilla chinesis, revealed significant anti-hyperglycemic, anti-hyperlipidemic and antioxidant activities.
本研究从委陵菜中分离并鉴定出具有降血糖、降血脂和抗氧化作用的主要化合物——trans-tiliroside。
从委陵菜乙醇提取物中分离出活性化合物 trans-tiliroside,并对其给药剂量进行优化和专利申请。采用正常小鼠、四氧嘧啶诱导的糖尿病小鼠和链脲佐菌素诱导的糖尿病大鼠来评价委陵菜 trans-tiliroside 的降血糖、降血脂和抗氧化作用。检测不同剂量 trans-tiliroside 处理的四氧嘧啶诱导的糖尿病小鼠和链脲佐菌素诱导的糖尿病大鼠的血液生化参数。
连续 15 天每天口服 0.4、0.8 和 1.6mg/kg/day 剂量的 trans-tiliroside,可显著降低四氧嘧啶诱导的糖尿病小鼠空腹血清葡萄糖、甘油三酯(TG)和总胆固醇(TC)水平。trans-tiliroside(1.2 和 0.3mg/kg 体重,10 周)可显著降低 STZ 诱导的糖尿病大鼠的血糖水平。TC、低密度脂蛋白胆固醇(LDL-C)和 TG 水平降低,高密度脂蛋白胆固醇(HDL-C)水平升高,改善了血脂代谢。此外,trans-tiliroside 还具有抗氧化活性,表现为糖尿病大鼠中超氧化物歧化酶(SOD)活性增加,丙二醛(MDA)水平降低。组织形态学检查表明,trans-tiliroside 可恢复糖尿病大鼠胰腺组织的损伤。
