Abstract

OBJECTIVE

To study the effect of genistein on the expression of heme oxygenase-1 (HO-1) in rats with pulmonary arterial hypertension (PAH) induced by monocrotaline (MCT).

METHODS

Sixty male Sprague-Dawley rats were randomly divided into 4 groups (n=15), namely the control group, model group, low-dose (20 µg/kg) genistein group and high-dose (80 µg/kg) genistein group. The hemodynamic parameters were measured and the remodeling of pulmonary small arteries was observed by electron microscope (EM). The expression of HO-1 in the lung tissues were detected by Western blotting.

RESULTS

Compared with the model group, genistein treatment significantly reduced the elevated mean pulmonary arterial pressure, improved the right ventricular hypertrophy index, and increased the expression of HO-1 in a dose-dependent manner.

CONCLUSION

Genistein attentuates pulmonary arterial hypertension in MCT-treated rats possibly by up-regulation of HO-1 in the lung tissues.

目的

研究染料木黄酮对野百合碱(MCT)诱导的肺动脉高压(PAH)大鼠血红素加氧酶-1(HO-1)表达的影响。

方法

将60只雄性Sprague-Dawley大鼠随机分为4组(n = 15),即对照组、模型组、低剂量(20μg/kg)染料木黄酮组和高剂量(80μg/kg)染料木黄酮组。测量血流动力学参数,并用电子显微镜(EM)观察肺小动脉的重塑情况。通过蛋白质印迹法检测肺组织中HO-1的表达。

结果

与模型组相比,染料木黄酮治疗显著降低了升高的平均肺动脉压,改善了右心室肥厚指数,并以剂量依赖的方式增加了HO-1的表达。

结论

染料木黄酮可能通过上调肺组织中HO-1的表达来减轻MCT处理大鼠的肺动脉高压。
