To test whether membrane fluidity and its changes are important for the sensitivity of cells to the action of perifosine (OPP), a new anticancer drug targeting cell membrane and not DNA.
Influence of OPP on the membrane structure of OPP-resistant MCF7, and OPP-sensitive MT3 breast cancer cell lines, as well as of mouse fibroblasts (L929) cell lines, and model cells (liposomes) was investigated by electron paramagnetic resonance, using spin labeled derivative of OPP (P5) and 5-doxylpalmitoyl methylester (MeFASL(10,3)) as spin probes.
OPP increased membrane fluidity of all cell lines at concentrations higher than 50 μM (on the level of P≤0.05, t test). In cells, the differences were observed only by P5 and not by MeFASL(10,3). Average order parameter Seff decreased for about 12% in MCF7 and L929 and only for 8% in OPP-sensitive MT3 cells, showing that there was no correlation between membrane fluidity changes and sensitivity of cells to OPP. The only correlation we found was between OPP sensitivity and the cell growth rate. In liposomes, both spin probes were sensitive to the action of OPP. Seff decreased with increasing concentration of OPP. For MeFASL(10,3) a significant decrease was observed at 4 mol% OPP, while for P5 it was observed at 8 mol%.
Influence of OPP on plasma membrane fluidity of breast cancer cells is not the determining factor in the sensitivity of cells to OPP.
检测膜流动性及其变化对于细胞对新型抗癌药物哌立福新(OPP)敏感性的重要性,该药物作用靶点为细胞膜而非DNA。
采用电子顺磁共振技术,以OPP的自旋标记衍生物(P5)和5-脱氧棕榈酰甲基酯(MeFASL(10,3))作为自旋探针,研究OPP对耐OPP的MCF7、对OPP敏感的MT3乳腺癌细胞系以及小鼠成纤维细胞(L929)细胞系和模型细胞(脂质体)膜结构的影响。
当浓度高于50μM时,OPP增加了所有细胞系的膜流动性(P≤0.05,t检验)。在细胞中,仅通过P5观察到差异,而MeFASL(10,3)未观察到差异。MCF7和L929细胞中平均序参数Seff下降约12%,而在对OPP敏感的MT3细胞中仅下降8%,表明膜流动性变化与细胞对OPP的敏感性之间无相关性。我们发现的唯一相关性是OPP敏感性与细胞生长速率之间的相关性。在脂质体中,两种自旋探针均对OPP的作用敏感。Seff随OPP浓度增加而下降。对于MeFASL(10,3),在4mol% OPP时观察到显著下降,而对于P5,在8mol%时观察到显著下降。
