Abstract

OBJECTIVE

To investigate the expression of A disintegrin and metalloproteinase 17 (ADAM17) mRNA and ADAM17 protein in esophageal squamous cell carcinoma (ESCC), and to evaluate their correlation with clinicopathological features and prognosis.

METHODS

The expression of ADAM17 mRNA in 50 ESCC and 50 normal esophageal tissues was detected by RT-PCR. The expression of ADAM17 protein in 80 ESCC and 80 normal esophageal tissues was detected with immunohistochemioal staining (SP). Log rank test and Cox proportional hazards analysis were used to analyze the prognosis of ESCC.

RESULTS

The expression of ADAM17 mRNA in 50 ESCC and 50 normal esophageal tissues was 0.937 ± 0.241 and 0.225 ± 0.077, respectively. The positive expression rates of ADAM17 protein in 80 ESCC and 80 normal esophageal tissues was 66.2% and 6.2%, respectively. The expressions of ADAM17 mRNA and ADAM17 protein in the ESCC were significantly higher than those in normal esophageal tissues (P < 0.01). The expressions of ADAM17 mRNA and protein were positively correlated with lymph node metastasis and TNM staging (P < 0.05). There were no correlations between the expressions of ADAM17 mRNA and protein and sex, age and histological grade (P > 0.05) . The expression of ADAM17 protein was positively correlated with EGFR protein (P < 0.01). The lymph node metastasis, TNM staging and the expression of ADAM17 and EGFR protein were independent prognostic factors.

CONCLUSIONS

ADAM17 mRNA and protein are highly expressed in ESCC than in normal esophageal tissues and may play an important role in the development, invasion and metastasis of ESCC. They may be used as prognostic factors of ESCC.

目的

探讨去整合素和金属蛋白酶17(ADAM17)mRNA及蛋白在食管鳞状细胞癌(ESCC)中的表达情况,并评估其与临床病理特征及预后的相关性。

方法

采用逆转录聚合酶链反应(RT-PCR)检测50例ESCC组织及50例正常食管组织中ADAM17 mRNA的表达。采用免疫组织化学染色(SP法)检测80例ESCC组织及80例正常食管组织中ADAM17蛋白的表达。采用对数秩检验和Cox比例风险分析评估ESCC的预后。

结果

50例ESCC组织及50例正常食管组织中ADAM17 mRNA的表达分别为0.937±0.241和0.225±0.077。80例ESCC组织及80例正常食管组织中ADAM17蛋白的阳性表达率分别为66.2%和6.2%。ESCC组织中ADAM17 mRNA及蛋白的表达均显著高于正常食管组织(P<0.01)。ADAM17 mRNA及蛋白的表达与淋巴结转移及TNM分期呈正相关(P<0.05)。ADAM17 mRNA及蛋白的表达与性别、年龄及组织学分级无相关性(P>0.05)。ADAM17蛋白的表达与表皮生长因子受体(EGFR)蛋白呈正相关(P<0.01)。淋巴结转移、TNM分期及ADAM17和EGFR蛋白的表达是独立的预后因素。

结论

与正常食管组织相比,ADAM17 mRNA及蛋白在ESCC中高表达,可能在ESCC的发生、侵袭和转移中起重要作用。它们可能作为ESCC的预后因素。
