Abstract

OBJECTIVE

To investigate the association of human leukocyte antigen-E (HLA-E) gene polymorphisms and plasma soluble HLA-E (sHLA-E) levels with genetic susceptibility to breast cancer in Zhangjiakou women of Han nationality.

METHODS

HLA-E typing was performed in 200 patients with breast cancer and 228 healthy controls from Zhangjiakou area by means of polymerase chain reaction-sequence-specific priming (PCR-SSP). Simultaneously, sHLA-E was detected by ELISA.

RESULTS

We found two alleles of HLA-E: HLA-E0101 and HLA-E0103, and three genotypes, namely HLA-E0101/HLA-E0101, HLA-E0101/HLA-E0103 and HLA-E0103/HLA-E0103. The frequencies of HLA-E0103 allele and HLA-E0103/HLA-E0103 genotype showed a significant difference between healthy controls and patients with breast cancer. The HLA-E0103/HLA-E0103 genotype significantly increased the risk for breast cancer. Plasma levels of sHLA-E showed no significant difference in breast cancer patients and controls. While in breast cancer patients with HLA-E0103/HLA-E*0103 genotype, plasma levels of sHLA-E were significantly elevated compared with those in healthy controls. HLA-E genotypes and sHLA-E levels had no obvious relationship with tumor grading.

CONCLUSION

HLA-E gene polymorphisms are associated with the development of breast cancer in Zhangjiakou women of Han nationality. HLA-E0103/HLA-E0103 genotype is probably susceptible genotype for breast cancer, while plasma sHLA-E level probably has no association with breast cancer susceptibility.

目的

探讨人类白细胞抗原-E(HLA-E)基因多态性及血浆可溶性HLA-E(sHLA-E)水平与张家口地区汉族女性乳腺癌遗传易感性的关系。

方法

采用聚合酶链反应-序列特异性引物法(PCR-SSP)对张家口地区200例乳腺癌患者及228例健康对照者进行HLA-E分型。同时,采用酶联免疫吸附测定法(ELISA)检测sHLA-E。

结果

发现HLA-E基因有两个等位基因:HLA-E0101和HLA-E0103,三种基因型,即HLA-E0101/HLA-E0101、HLA-E0101/HLA-E0103和HLA-E0103/HLA-E0103。HLA-E0103等位基因频率及HLA-E0103/HLA-E0103基因型频率在健康对照者与乳腺癌患者之间存在显著差异。HLA-E0103/HLA-E0103基因型显著增加乳腺癌发病风险。乳腺癌患者与对照者血浆sHLA-E水平无显著差异。而HLA-E0103/HLA-E*0103基因型的乳腺癌患者,其血浆sHLA-E水平较健康对照者显著升高。HLA-E基因型及sHLA-E水平与肿瘤分级无明显关系。

结论

HLA-E基因多态性与张家口地区汉族女性乳腺癌的发生有关。HLA-E0103/HLA-E0103基因型可能是乳腺癌的易感基因型,而血浆sHLA-E水平可能与乳腺癌易感性无关。
