Abstract

OBJECTIVE

Methotrexate (MTX) leads to acute toxic side effects in tissues or organs containing rapidly dividing cells such as seminiferous tubules. In this study, we investigated the protective effects of vitamin C against MTX-induced injury in rat testis.

MATERIAL AND METHODS

A total of 31 rats were divided into 4 groups, including the control group. The study was completed within 4 weeks and the rats received daily doses of 2 mL/kg SF, 100 mg/kg vitamin C and 10 mg/kg/day MTX i.p according to their groups. The mean seminiferous tubular diameter, germinal epithelial cell thickness, and mean testicular biopsy score were determined by histologic examination of each group.

RESULTS

The vitamin C + MTX group showed more similarity with the control group. Statistically significant results were achieved between groups as for mean seminiferous tubular diameter, germinal epithelial cell thickness, and mean testicular biopsy score. When compared with the group which received vitamin C after MTX therapy, values for mean seminiferous tubular diameter, germinal epithelial cell thickness, and mean testicular biopsy score were significantly higher in the group which received vitamin C before initiation of MTX therapy.

CONCLUSION

Vitamin C decreased MTX-induced testicular histological injuries, especially when used before MTX therapy.

目的

甲氨蝶呤(MTX)会对含有快速分裂细胞的组织或器官(如曲细精管)产生急性毒性副作用。在本研究中,我们调查了维生素C对MTX诱导的大鼠睾丸损伤的保护作用。

材料与方法

总共31只大鼠被分为4组,包括对照组。研究在4周内完成,大鼠根据分组每天腹腔注射2 mL/kg生理盐水、100 mg/kg维生素C和10 mg/kg/天MTX。通过对每组进行组织学检查来测定曲细精管平均直径、生精上皮细胞厚度和平均睾丸活检评分。

结果

维生素C + MTX组与对照组表现出更多相似性。在曲细精管平均直径、生精上皮细胞厚度和平均睾丸活检评分方面,各组之间取得了具有统计学意义的结果。与MTX治疗后接受维生素C的组相比,在MTX治疗开始前接受维生素C的组中,曲细精管平均直径、生精上皮细胞厚度和平均睾丸活检评分的值显著更高。

结论

维生素C减轻了MTX诱导的睾丸组织学损伤,尤其是在MTX治疗前使用时。
