To evaluate the safety of high-intensity focused ultrasound keratoplasty as a treatment for presbyopia by examining its effect on the rabbit anterior segment. The right corneas of 36 New Zealand rabbits were treated with HIFU keratoplasty. The animals were sacrificed at 1, 7, 15, 30, 60, and 90 days after operation. Collagen type I, MMP-2, and MMP-9 were evaluated using immunohistochemistry. For the detection of apoptosis, the TUNEL method was applied. The SOD and MDA levels were analyzed with assay kits. Collagen type I, MMP-2, and MMP-9 levels were altered after the operation but returned to normal within 90 days. The apoptotic index (AI) of the corneal cells decreased from 1 to 30 days gradually. No apoptosis was observed in the epithelial cells of the lens, and the SOD and MDA levels were normal at any time point. After HIFU keratoplasty, the histomorphology of the cornea changed, the corneal collagen type I levels decreased, the corneal MMP-2 and MMP-9 levels increased, and the corneal cells underwent apoptosis for a period of time. Ninety days after the operation, the levels returned to normal, and the lenses were not affected. Thus, HIFU presents good biological safety for eyes.
通过检查高强度聚焦超声角膜移植术对兔眼前节的影响,评估其作为治疗老花眼方法的安全性。对36只新西兰兔的右眼进行高强度聚焦超声角膜移植术治疗。在术后1、7、15、30、60和90天处死动物。采用免疫组织化学法评估Ⅰ型胶原、基质金属蛋白酶-2(MMP-2)和基质金属蛋白酶-9(MMP-9)。应用TUNEL法检测细胞凋亡。用试剂盒分析超氧化物歧化酶(SOD)和丙二醛(MDA)水平。术后Ⅰ型胶原、MMP-2和MMP-9水平发生改变,但在90天内恢复正常。角膜细胞凋亡指数(AI)在1至30天逐渐下降。晶状体上皮细胞未观察到凋亡,且在任何时间点SOD和MDA水平均正常。高强度聚焦超声角膜移植术后,角膜组织形态学发生改变,角膜Ⅰ型胶原水平降低,角膜MMP-2和MMP-9水平升高,角膜细胞在一段时间内发生凋亡。术后90天,各项指标恢复正常,晶状体未受影响。因此,高强度聚焦超声对眼睛具有良好的生物安全性。
