Abstract

AIM

α-Synuclein (aSyn), a putative cerebrospinal fluid biomarker, may support the diagnosis of neurodegenerative diseases. Previous studies led to conflicting results due to different preanalytical and analytical procedures. Standardized assays are required to allow for comparison of results from different laboratories.

MATERIALS & METHODS: We performed a side-by-side validation of a commercially available (MSD, MD, USA) and a 'homebrew' assay for quantification of aSyn according to published guidelines.

RESULTS

The data showed high sensitivity and reproducibility for both assays. Preanalytical and analytical parameters did not affect the outcome of measurements.

CONCLUSION

We conclude that both assays are very close in performance and suitable for research application of cerebrospinal fluid aSyn.

目的

α-突触核蛋白(aSyn)作为一种假定的脑脊液生物标志物,可能有助于神经退行性疾病的诊断。由于不同的分析前和分析程序,先前的研究结果相互矛盾。需要标准化的检测方法以便比较不同实验室的结果。

材料与方法

我们根据已发表的指南,对市售的(美国马里兰州MSD公司)和一种“自制”的用于定量aSyn的检测方法进行了并行验证。

结果

数据显示两种检测方法均具有高灵敏度和可重复性。分析前和分析参数不影响测量结果。

结论

我们得出结论,两种检测方法在性能上非常接近,适用于脑脊液aSyn的研究应用。
