Abstract

PURPOSE

To evaluate the performance of the xTAG liquid chip technology (xTAG-LCT) for etiological diagnosis of acute retinal necrosis (ARN).

METHODS

Fifteen vitreous and 3 aqueous samples from 18 ARN patients were analyzed by xTAG-LCT and multiplex PCR (mPCR)/quantitative PCR (qPCR).

RESULTS

xTAG-LCT revealed positive results in 17 of the 18 samples: 10 for Varicella Zoster Virus (VZV) alone; 5 for VZV and Epstein-Barr virus (EBV); 1 for herpes simplex viruses type 1 (HSV-1) and EBV; 1 for VZV, HSV-1 and EBV. While mPCR revealed the same results as xTAG-LCT for VZV and HSV-1 in all samples, only 2 of the 7 samples positive for EBV on xTAG-LCT were confirmed by qPCR. None of the 28 control vitreous samples from 8 non-ARN patients and 10 pair of cadaveric eyes was positive for any of the tested viruses.

CONCLUSIONS

xTAG-LCT could be a useful alternative for etiological diagnosis of ARN.

评估 xTAG 液相芯片技术(xTAG-LCT)在急性视网膜坏死(ARN)病因诊断中的性能。

方法

对 18 例 ARN 患者的 15 份玻璃体和 3 份房水样本进行了 xTAG-LCT 和多重聚合酶链反应(mPCR)/定量聚合酶链反应(qPCR)分析。

结果

xTAG-LCT 在 18 例样本中的 17 例中显示出阳性结果:单纯水痘带状疱疹病毒(VZV)10 例;VZV 和 EBV 5 例;单纯单纯疱疹病毒 1 型(HSV-1)和 EBV 1 例;VZV、HSV-1 和 EBV 各 1 例。虽然 mPCR 在所有样本中均与 xTAG-LCT 对 VZV 和 HSV-1 显示出相同的结果,但 xTAG-LCT 对 EBV 呈阳性的 7 个样本中,仅通过 qPCR 证实了 2 个样本。8 例非 ARN 患者和 10 对尸眼的 28 例对照玻璃体样本均未对任何检测到的病毒呈阳性。
