Abstract

OBJECTIVE

To discover the expression pattern and potential underlying mechanism of the caspase recruitment domain-containing protein 9 (CARD9) in oral squamous cell carcinoma (OSCC).

METHODS

Caspase recruitment domain-containing protein 9 expression was detected by qRT-PCR and Western blot in OSCC tissues and cells, and OSCC (CGHNC9 and OECM-1) cell lines were divided into control, NC siRNA, and CARD9 siRNA groups. Then, MTT, flow cytometry, wound-healing, and Transwell assays were carried out to determine the changes in cellular biological characteristics. Immunoblot assay was performed for the expressions of NF-κB pathway. Finally, we constructed the xenograft models in nude mice to validate the in vivo effect of CARD9 siRNA on OSCC cell growth.

RESULTS

Caspase recruitment domain-containing protein 9 was upregulated in both OSCC tissues and cells, exhibiting a close relation with major clinicopathological features of OSCC patients. Transfection of CARD9 siRNA inhibited the proliferation, migration, and invasion of OSCC cells with the enhanced cell apoptosis, and meanwhile, CARD9, p-p65/p65, p-IKKα/IKKα, and p-IkBα/IkBα were downregulated. The tumor formation assay on nude mice also suggested that CARD9 siRNA might block the in vivo growth of OSCC cells.

CONCLUSION

Caspase recruitment domain-containing protein 9 suppression results in the upregulation of NF-κB pathway with suppressed proliferation, migration, and invasion of OSCC cells and facilitates the apoptosis.

目的

探索衔接蛋白 caspase 募集域蛋白 9(CARD9)在口腔鳞状细胞癌(OSCC)中的表达模式及其潜在的作用机制。

方法

采用 qRT-PCR 和 Western blot 检测 OSCC 组织和细胞中 CARD9 的表达,将 OSCC(CGHNC9 和 OECM-1)细胞系分为对照组、阴性对照(NC)siRNA 组和 CARD9 siRNA 组,分别采用 MTT、流式细胞术、划痕愈合实验和 Transwell 实验检测细胞生物学特性的变化,免疫印迹法检测 NF-κB 通路的表达,最后构建裸鼠异种移植模型,验证 CARD9 siRNA 对 OSCC 细胞生长的体内作用。

结果

CARD9 在 OSCC 组织和细胞中均呈高表达,与 OSCC 患者的主要临床病理特征密切相关。转染 CARD9 siRNA 可抑制 OSCC 细胞的增殖、迁移和侵袭,促进细胞凋亡,同时下调 CARD9、p-p65/p65、p-IKKα/IKKα 和 p-IkBα/IkBα。裸鼠成瘤实验也表明,CARD9 siRNA 可能阻断 OSCC 细胞的体内生长。

结论

CARD9 抑制可导致 NF-κB 通路的上调,抑制 OSCC 细胞的增殖、迁移和侵袭,并促进细胞凋亡。
