Abstract

OBJECTIVE

To investigate the effects of lycopene on metabolism of glycolipid and inflammation in rats with non-alcoholic fatty liver disease.

METHODS

According to body weight, sixty male Sprague-Dawley rats were randomly divided into control group, non-alcoholic fatty liver model group, 20, 60 mg/kg lycopene intervention groups, with 15 rats in each group. The control group was fed normal diet, and the other 3 groups were fed high-fat and high-fructose diet for 4 weeks to establish the model of non-alcoholic fatty liver disease. After modeling, the intervention groups were fed lycopene of different doses by gavage. After 8 weeks of continuous feeding, the rats were sacrificed, the body weight and liver weight were weighed, and the liver index was calculated. Hematoxylin-eosin staining was used to observe the morphological changes of liver tissue. Serum was collected, fasting blood glucose(FBG), fasting insulin(INS), and insulin resistance index(HOMA-IR) were calculated; Liver function indicators alanine aminotransferase(ALT), aspartate aminotransferase(AST) were measured; Serum triglycerides(TG), total cholesterol(TC), high density lipoprotein-cholesterol(HDL-C), low density lipoprotein-cholesterol(LDL-C) and levels of inflammatory cytokines interleukin-6(IL-6), interleukin-18(IL-18) and interleukin-1β(IL-1β) were measured.

RESULTS

Compared with the control group, the liver weight and liver index in the model group increased by 27% and 24%, respectively; And steatosis occurred in the liver tissue; The levels of serum ALT, TG, TC, LDL-C, IL-6, IL-1β were significantly increased, the level of serum HDL-C was significantly decreased(P<0. 05). Compared with the model group, liver weight, liver index, levels of serum ALT, TG, FBG, IL-6 and IL-1β were significantly lower in the 20, 60 mg/kg lycopene intervention groups(P<0. 05), the INS and HOMA-IR index showed a downward trend; Liver tissue lesions were reduced to different degrees, and the effect was more significant in the 60 mg/kg lycopene group.

CONCLUSION

Lycopene can improve non-alcoholic fatty liver disease by regulating glycolipid metabolism and reducing levels of inflammatory cytokines in rats.

目的

探讨番茄红素对非酒精性脂肪性肝病大鼠糖脂代谢及炎症的影响。

方法

将60只雄性Sprague-Dawley大鼠按体重随机分为对照组、非酒精性脂肪肝模型组、20、60mg/kg番茄红素干预组,每组15只。对照组给予正常饮食,其余3组给予高脂高糖饮食4周以建立非酒精性脂肪性肝病模型。造模后,干预组通过灌胃给予不同剂量的番茄红素。连续喂养8周后,处死大鼠,称取体重和肝脏重量,计算肝脏指数。采用苏木精-伊红染色观察肝组织形态学变化。采集血清,计算空腹血糖(FBG)、空腹胰岛素(INS)及胰岛素抵抗指数(HOMA-IR);检测肝功能指标丙氨酸氨基转移酶(ALT)、天冬氨酸氨基转移酶(AST);测定血清甘油三酯(TG)、总胆固醇(TC)、高密度脂蛋白胆固醇(HDL-C)、低密度脂蛋白胆固醇(LDL-C)及炎症细胞因子白细胞介素-6(IL-6)、白细胞介素-18(IL-18)和白细胞介素-1β(IL-1β)水平。

结果

与对照组相比,模型组肝脏重量和肝脏指数分别增加27%和24%;肝组织出现脂肪变性;血清ALT、TG、TC、LDL-C、IL-6、IL-1β水平显著升高,血清HDL-C水平显著降低(P<0.05)。与模型组相比,20、60mg/kg番茄红素干预组肝脏重量、肝脏指数、血清ALT、TG、FBG、IL-6和IL-1β水平显著降低(P<0.05),INS及HOMA-IR指数呈下降趋势;肝组织病变不同程度减轻,60mg/kg番茄红素组效果更显著。

结论

番茄红素可通过调节大鼠糖脂代谢及降低炎症细胞因子水平改善非酒精性脂肪性肝病。
