Infections of Cyprinid herpesvirus 2 in goldfish and farmed crucian carp (Carassius auratus gibelio) are still an urgent problem worldwide. Detection and prevention are necessary for the control of haematopoietic necrosis disease caused by CyHV-2. Although many sensitive molecular diagnostic methods have been developed, effective immunodiagnosis and neutralization approaches based on monoclonal antibodies (MAbs) against CyHV-2 are still important to CyHV-2 study. In this experiment, purified CyHV-2 was used as antigens to produce monoclonal antibodies (Mabs). Six Mabs bound to different proteins were selected by Dot-blot screening and Western-blot analysis, and no one had cross-reactivity with closely related koi herpesvirus. Among them, Mabs 2E1-B10, 1F5-A3 and 4C4-A7 belonged to IgG isotype, while other three Mabs 3G9-B11, 3B4-G5 and 4F4-B7 belonged to IgM isotype. These six Mabs all could specifically detect CyHV-2 in CyHV-2 infected caudal fin of Carassius auratus gibelio (GiCF) cells by immunofluorescence assays. Then, the neutralization ability was tested in vitro, and the result showed that all six Mabs can attenuate CPE by CyHV-2 in vitro among which 2E1-B10 had the best neutralization ability. The virus proteins recognized by these six Mabs were identified by mass spectrometry identification, and the result showed they probably were ORF88, ORF55R, ORF115 and ORF151R. This study is the first to prepare Mabs by purifying CyHV-2, which will provide a practical basis for the in-depth study of CyHV-2 virus.
鲤鱼疱疹病毒 2 (Cyprinid herpesvirus 2,CyHV-2)感染的金鱼和养殖鲫鱼(Carassius auratus gibelio)仍然是全球范围内的一个紧迫问题。为了控制由 CyHV-2 引起的造血坏死病,检测和预防是必要的。虽然已经开发出许多敏感的分子诊断方法,但基于单克隆抗体(Monoclonal antibodies,MAbs)的有效免疫诊断和中和方法对于 CyHV-2 的研究仍然很重要。在本实验中,使用纯化的 CyHV-2 作为抗原来制备单克隆抗体(MAbs)。通过 Dot-blot 筛选和 Western-blot 分析,选择了 6 株与不同蛋白结合的 MAbs,并且它们都与密切相关的锦鲤疱疹病毒无交叉反应。其中,Mabs 2E1-B10、1F5-A3 和 4C4-A7 属于 IgG 同种型,而其他 3 株 Mabs 3G9-B11、3B4-G5 和 4F4-B7 属于 IgM 同种型。这些 MAbs 均能通过免疫荧光检测特异性检测感染鲫鱼尾鳍细胞(Carassius auratus gibelio,GiCF)的 CyHV-2。然后,在体外测试了中和能力,结果表明,这 6 株 MAbs 均能在体外中和 CyHV-2 引起的细胞病变效应(Cytopathic effect,CPE),其中 2E1-B10 的中和能力最强。通过质谱鉴定识别出这些 MAbs 识别的病毒蛋白,结果表明它们可能是 ORF88、ORF55R、ORF115 和 ORF151R。本研究首次通过纯化 CyHV-2 制备 MAbs,为深入研究 CyHV-2 病毒提供了实用基础。
