The plasma membrane calcium pump (PMCA) is an important transporter that maintains intracellular calcium concentration ([Ca]). It allows the calcium (Ca) from inside the cell to go out of the cell through the plasma membrane. For this, it cooperates with the proteins in the cell. The aim of this study is to demonstrate the effect of PMCA on intracellular calcium signaling in breast cancer cells. In this study, PMCA was inhibited by orthovanadate (OV), and changes in Calmodulin (CaM), Calcineurin (CaN) and cMyc proteins were demonstrated. Intracellular calcium accumulation was measured when PMCA was inhibited in MDA-MB-231 cells. At the same time, it was observed that the cell movement decreased with time. Over time, CaN and CaM were slightly suppressed, and cMyc protein was not expressed. As a result, when PMCA protein is targeted correctly in breast cancer cells, it has an indirect effect on cancer-promoting proteins.
质膜钙泵(PMCA)是一种维持细胞内钙浓度([Ca])的重要转运蛋白。它使细胞内的钙(Ca)通过质膜排出细胞。为此,它与细胞内的蛋白质协同作用。本研究的目的是证明PMCA对乳腺癌细胞内钙信号传导的影响。在本研究中,原钒酸盐(OV)抑制了PMCA,并证明了钙调蛋白(CaM)、钙调神经磷酸酶(CaN)和cMyc蛋白的变化。在MDA-MB-231细胞中抑制PMCA时,测量细胞内钙积累。同时,观察到细胞运动随时间减少。随着时间的推移,CaN和CaM受到轻微抑制,cMyc蛋白未表达。结果,当在乳腺癌细胞中正确靶向PMCA蛋白时,它对促癌蛋白有间接影响。
