This study aimed to evaluate the effects of topical application of methanolic extract on signal transducer and activator of transcription 3 (STAT3) expression and tongue epithelial cell death caused by oral candidiasis under immunosuppressive conditions.
To obtain the oral candidiasis model, 20 healthy male (Wistar) rats were administered oral dexamethasone and tetracycline for 14 days and oral candidiasis was induced with (ATCC-10231) 1 McFarland. They were then randomized into four groups-immunosuppression (K-), oral candidiasis (K + ), nystatin treatment (P1), and 20% methanolic extract treatment (P2) and were treated for 14 days. Histological analyses of cell death and candida invasion and immunohistochemical analysis of STAT3 in epithelial cells were performed.
Epithelial cell death data were analyzed using one-way analysis of variance (ANOVA) and the post hoc Games-Howell test ( < 0.05) and STAT3 expression with one-way ANOVA and the post hoc least significant difference test ( < 0.05).
Cell death was significantly different between K- and K+ and between K+ and P1 and P2 ( < 0.05); there were no significant differences between K- and P1 and P2 and between P1 and P2 ( > 0.05). STAT3 expression was significantly different between K- and P1 and P2 and between K+ and P1 and P2 ( < 0.05), but there were no significant differences between K+ and K- and between P1 and P2 ( > 0.05).
Topical administration of methanol extract increased STAT3 expression and decreased tongue epithelial cell death caused by oral candidiasis.
本研究旨在评估在免疫抑制条件下,局部应用甲醇提取物对口腔念珠菌病所致信号转导和转录激活因子3(STAT3)表达及舌上皮细胞死亡的影响。
为建立口腔念珠菌病模型,给20只健康雄性(Wistar)大鼠口服地塞米松和四环素14天,并用1麦氏浊度的白色念珠菌(ATCC - 10231)诱导口腔念珠菌病。然后将它们随机分为四组——免疫抑制组(K-)、口腔念珠菌病组(K +)、制霉菌素治疗组(P1)和20%甲醇提取物治疗组(P2),并治疗14天。进行细胞死亡和念珠菌侵袭的组织学分析以及上皮细胞中STAT3的免疫组织化学分析。
使用单因素方差分析(ANOVA)和事后检验Games - Howell检验(P < 0.05)分析上皮细胞死亡数据,使用单因素方差分析和事后检验最小显著差异检验(P < 0.05)分析STAT3表达。
K-组与K +组之间以及K +组与P1组和P2组之间的细胞死亡存在显著差异(P < 0.05);K-组与P1组和P2组之间以及P1组与P2组之间无显著差异(P > 0.05)。K-组与P1组和P2组之间以及K +组与P1组和P2组之间的STAT3表达存在显著差异(P < 0.05),但K +组与K-组之间以及P1组与P2组之间无显著差异(P > 0.05)。
局部应用甲醇提取物可增加STAT3表达,并减少口腔念珠菌病所致的舌上皮细胞死亡。
