Idiotypes (Id) are unique antigenic determinants in hypervariable regions of immunoglobulins. This paper describes the clinical usefulness of Id for the determination of pathogenic autoantibodies. O-81-Id, human anti-DNA antibody idiotypes, were specifically detected in circulating and renal glomeruli-deposited immune complexes of patients with active lupus nephritis. Anti-idiotypic antibodies to anti-DNA antibodies could be also detected in human sera. Anti-Id antibodies to O-81-Id occurred spontaneously during the remission of systemic lupus erythematosus (SLE) but rarely during an acute episode. Monoclonal anti-Id antibodies had an ability to suppress in vitro anti-DNA antibody production by SLE lymphocytes. We also presented specific manipulation of anti-DNA production though the elimination of anti-DNA-Id-positive cells by using anti-Id-cytotoxic agent conjugates.
独特型(Id)是免疫球蛋白高变区中独特的抗原决定簇。本文描述了独特型在致病性自身抗体测定中的临床应用。O-81-Id,即人抗DNA抗体独特型,在活动性狼疮性肾炎患者的循环免疫复合物和肾小球沉积免疫复合物中被特异性检测到。在人血清中也能检测到抗DNA抗体的抗独特型抗体。针对O-81-Id的抗独特型抗体在系统性红斑狼疮(SLE)缓解期自发出现,但在急性发作期很少出现。单克隆抗独特型抗体具有抑制SLE淋巴细胞体外抗DNA抗体产生的能力。我们还通过使用抗独特型细胞毒性剂偶联物消除抗DNA-Id阳性细胞,展示了对抗DNA产生的特异性调控。
