Atomic force microscopy was used to visualize the cruciform structure in supercoiled plasmid pUC8 DNA immobilized on aminomodified mica. The cruciform hairpin was 14 base pairs in size, as determined from atomic force microscopy images of pUC8 DNA in air. Molecular modeling confirmed that the cruciform structure is formed by hairpins with self-complementary homopyrimidine-homopurine sequences (dT)8(dA)6 and a loop 4 nucleotides long.
原子力显微镜用于观察固定在氨基修饰云母上的超螺旋质粒pUC8 DNA中的十字形结构。从空气中pUC8 DNA的原子力显微镜图像确定,十字形发夹大小为14个碱基对。分子建模证实,十字形结构由具有自我互补同嘧啶-同嘌呤序列(dT)8(dA)6和一个4个核苷酸长环的发夹形成。
