The aim of this study was to determine the effects of bilirubin in experimental small intestinal ischemia/reperfusion (I/R) injury in rats.
Thirty rats were divided into 5 groups (n = 6). In group S, saline and in group B, bilirubin, 20 mg/kg were infused via the jugular vein without an additional procedure. In groups S-IR, saline, B(1)-IR and B(2)-IR, 10 and 20 mg/kg/h of bilirubin were infused for 2 hours, respectively. In these groups, an I/R procedure was done after infusions by occluding the superior mesenteric artery for 45 minutes followed by 1 hour of reperfusion. After reperfusion, the small intestines were resected for histopathologic and malondialdehyde (MDA) assessments. Mucosal lesions were scored between 0 and 5. Malondialdehyde levels and histopathologic grades were analyzed statistically.
Mucosal injury was severe in S-IR (grade 4 to 5), mild in B(1)-IR (grade 0 to 3) and none in B(2)-IR group (grade 0). Grades of group S-IR were higher than those of B(1)-IR and B(2)-IR statistically (P <.05). Tissue MDA levels of the S-IR group were significantly higher than those of B(1)-IR and B(2)-IR groups (U = 36, P <.05). Bilirubin levels correlated inversely with MDA levels (r = -0.94).
Bilirubin effectively prevents intestinal I/R injury in rat. This observation is consistent with the hypotheses regarding bilirubin as an antioxidant, having a role in the body defense. J Pediatr Surg 36:1764-1767.
本研究旨在确定胆红素对大鼠实验性小肠缺血/再灌注(I/R)损伤的影响。
30只大鼠分为5组(每组n = 6)。S组经颈静脉输注生理盐水,B组经颈静脉输注20 mg/kg胆红素,均无额外操作。S-IR组、B(1)-IR组和B(2)-IR组分别以10 mg/kg/h和20 mg/kg/h的速度输注胆红素2小时。在这些组中,输注后通过阻断肠系膜上动脉45分钟,然后再灌注1小时来进行I/R操作。再灌注后,切除小肠进行组织病理学和丙二醛(MDA)评估。黏膜损伤评分为0至5分。对丙二醛水平和组织病理学分级进行统计学分析。
S-IR组黏膜损伤严重(4至5级),B(1)-IR组轻度(0至3级),B(2)-IR组无损伤(0级)。S-IR组的分级在统计学上高于B(1)-IR组和B(2)-IR组(P <.05)。S-IR组的组织MDA水平显著高于B(1)-IR组和B(2)-IR组(U = 36,P <.05)。胆红素水平与MDA水平呈负相关(r = -0.94)。
