To investigate the significance and the influence of transforming growth factor beta 1 (TGF beta 1) on fibroblast proliferation and collagen synthesis.
The fibroblasts derived from healthy human dermis and pathologic scar were harvested and cultured in vitro with TGF beta 1. The effects of TGF beta 1 on the proliferation and collagen synthesis of the fibroblasts were investigated during the early phase (24 h) of cell confluence by means of nonradioactive cellular proliferation detection, 3H thymidine and 3H proline incorporation and DNA quantification analysis.
After the co-culture with TGF beta 1, the incorporation of 3H thymidine and 3H proline and the proliferation were enhanced significantly in the fibroblasts from pathologic scar (P < 0.01) but remained at normal level in those from healthy human dermis(P > 0.05).
The cell proliferation and collagen synthesis of scar-derived fibroblasts could be positively modulated and controlled by TGF beta 1, which might be very important in the scar formation.
探讨转化生长因子β1(TGFβ1)对成纤维细胞增殖及胶原合成的意义和影响。
取健康人真皮及病理性瘢痕来源的成纤维细胞,体外培养并加入TGFβ1。通过非放射性细胞增殖检测、3H-胸腺嘧啶核苷和3H-脯氨酸掺入及DNA定量分析,研究TGFβ1在细胞汇合早期(24小时)对成纤维细胞增殖及胶原合成的影响。
与TGFβ1共培养后,病理性瘢痕来源的成纤维细胞中3H-胸腺嘧啶核苷和3H-脯氨酸掺入及增殖显著增强(P<0.01),而健康人真皮来源的成纤维细胞则维持在正常水平(P>0.05)。
TGFβ1可正向调节和控制瘢痕来源成纤维细胞的细胞增殖及胶原合成,这在瘢痕形成中可能非常重要。
