This paper is aimed to explore the mechanisms of brain development abnormality induced by alcohol.
Astrocytes and oligodendrocytes of 19-day rat embryo were exploited and cultured in vitro, and alcohol and its metabolite product (acetaldehyde) were added to DMEMF(12) medium. After different exposure times, c-fos expression of astrocytes and oligodendrocytes was measured by the immunocytochemistry technique.
Changes in c-fos gene expression induced by alcohol and acetaldehyde was time and dose dependent. After 1 hr exposure, alcohol and acetaldehyde affected c-fos gene expression in two kinds of neuralglia. C-fos positive expression reached peak value after 2 hr, but recovered after 72 hr and showed special time phase expression.
Alcohol and acetaldehyde cause abnormal increase of c-fos gene expression in astrocytes and oligodendrocytes. This abnormal expression may play an important role in abnormal brain development induced by alcohol.
本文旨在探讨酒精诱导脑发育异常的机制。
利用19日龄大鼠胚胎的星形胶质细胞和少突胶质细胞进行体外培养,并将酒精及其代谢产物(乙醛)添加到DMEMF(12)培养基中。在不同暴露时间后,采用免疫细胞化学技术检测星形胶质细胞和少突胶质细胞的c-fos表达。
酒精和乙醛诱导的c-fos基因表达变化具有时间和剂量依赖性。暴露1小时后,酒精和乙醛影响两种神经胶质细胞中的c-fos基因表达。c-fos阳性表达在2小时后达到峰值,但在72小时后恢复,并呈现特殊的时间阶段表达。
酒精和乙醛导致星形胶质细胞和少突胶质细胞中c-fos基因表达异常增加。这种异常表达可能在酒精诱导的脑发育异常中起重要作用。
