The kinetics of 5-fluoruracil concentration in blood and urine in 18 patients with cancer of the pancreas was determined in a regional and systemic method of its administration. The concentration of 5-fluoruracil in blood was found to be, as a rule, lower and the time of detection shorted after i/a (regional) administration as compared with i/i (systemic) one. Accordingly, less amount of the substance involved is excreted with the urine in regional chemotherapy. This allows an application of large single and sessional doses in insignificant general toxic effects and also gaining much better therapeutic effect.
采用区域给药和全身给药方法,测定了18例胰腺癌患者血液和尿液中5-氟尿嘧啶浓度的动力学。结果发现,与静脉注射(全身给药)相比,动脉注射(区域给药)后血液中5-氟尿嘧啶的浓度通常较低,检测时间较短。因此,区域化疗时尿液中排出的该物质较少。这使得可以应用大剂量单次和阶段性给药,而全身毒性作用不明显,同时还能获得更好的治疗效果。
