The zona pellucida is a feasible candidate for contraceptive vaccine. Our previous study showed that a human zona pellucida (ZPA) recombinant protein produced antiserum that inhibited human in vitro fertilization in rabbits. The objective of this study is to examine whether the human recombinant protein produce fertilization-blocking antiserum in a non-human primate.
The amino terminal regions consisting of pig (198 amino acids) and human (206 amino acids) zona pellucida proteins were prepared by Escherichia coli to produce antisera in bonnet monkeys (Macaca radiata). Antibody production was assessed by immunofluorescent staining and enzyme-linked immunosorbent assay (ELISA). Fertilization-blocking test was carried out using human oocytes that had failed in intracytoplasmic sperm injection (ICSI) treatment under patient's informed consent.
Both antisera against pig and human recombinant proteins recognized native human zona pellucida by immunofluorescent staining. However, anti-recombinant human ZPA reacted to native pig zona protein more strongly than pig zona pellucida by ELISA. When human oocytes were treated with antisera before insemination, anti-human ZPA antiserum inhibited human sperm binding to human zona pellucida, but anti-pig ZPA antiserum did not.
Bonnet monkey produced fertilization-blocking antibody by immunization with human recombinant ZPA. The homologous or highly similar sequence is effective for developing a contraceptive vaccine when using recombinant peptides.
透明带是一种可行的避孕疫苗候选物。我们之前的研究表明,一种人透明带(ZPA)重组蛋白产生的抗血清可抑制兔体内的人类体外受精。本研究的目的是检测这种人重组蛋白是否能在非人灵长类动物中产生阻止受精的抗血清。
由大肠杆菌制备包含猪(198个氨基酸)和人(206个氨基酸)透明带蛋白的氨基末端区域,以在帽猴(食蟹猴)中产生抗血清。通过免疫荧光染色和酶联免疫吸附测定(ELISA)评估抗体产生情况。在患者知情同意的情况下,使用胞浆内单精子注射(ICSI)治疗失败的人类卵母细胞进行受精阻断试验。
通过免疫荧光染色,针对猪和人重组蛋白的两种抗血清均能识别天然人透明带。然而,通过ELISA检测,抗重组人ZPA对天然猪透明带蛋白的反应比对猪透明带更强。当在授精前用人抗血清处理人类卵母细胞时,抗人ZPA抗血清可抑制人类精子与人类透明带的结合,但抗猪ZPA抗血清则不能。
帽猴通过用人重组ZPA免疫产生了阻止受精的抗体。当使用重组肽时,同源或高度相似的序列对开发避孕疫苗有效。
