In order to improve the routine culture method for vascular endothelial cells, endothelial cells of vein from newborn umbilical cord were successfully isolated with combined using 0.1% collagenase and 0.25% trypsin (1:1). The endothelial cells were further abrated by mechanical method. The composition of culture medium was simplified by not adding vascular endothelial cell growth factor and heparin. The cultured cells were verified as vascular endothelial cells by morphology, ultramicromorphology and immunohistochemistry.
为改进血管内皮细胞的常规培养方法,采用0.1%胶原酶和0.25%胰蛋白酶(1:1)联合使用的方法成功分离出新生儿脐带静脉的内皮细胞。通过机械方法进一步刮除内皮细胞。通过不添加血管内皮细胞生长因子和肝素简化培养基成分。通过形态学、超微形态学和免疫组织化学鉴定培养的细胞为血管内皮细胞。
