To detect the bcr/abl fusion gene in chronic myeloid leukemia (CML) for assisting in clinical diagnosis.
Fluorescence in situ hybridization (FISH) and nested reverse transcription polymerase chain reaction (RT-PCR) were used.
Out of 16 CML patients, the results of FISH was not consistent with that of nested RT-PCR in 3 cases. When compared with the results of Northern blot, it was found that nested RT-PCR was more sensitive than FISH, but might give false positive results. Moreover, FISH was the easier method for detecting rarer types of bcr/abl transcripts. For detecting the latter occasions nested RT-PCR have to design several specific primers.
It could take their advantages of applying FISH and nested RT-PCR at the same time to make the results more accurate and reliable for the diagnosis, prognosis and monitor of minimal residual disease.
检测慢性髓性白血病(CML)中的bcr/abl融合基因以辅助临床诊断。
采用荧光原位杂交(FISH)和巢式逆转录聚合酶链反应(RT-PCR)。
16例CML患者中,3例FISH结果与巢式RT-PCR结果不一致。与Northern印迹结果比较发现,巢式RT-PCR比FISH更敏感,但可能出现假阳性结果。此外,FISH是检测罕见类型bcr/abl转录本的更简便方法。对于后一种情况,巢式RT-PCR必须设计几种特异性引物。
同时应用FISH和巢式RT-PCR的优势,可使结果在CML诊断、预后及微小残留病监测中更准确可靠。
