Abstract

OBJECTIVES

To investigate the role of endogenous heme oxygenase (HO)/carbon monoxide (CO) system in regulating the process of intussusception (IN) induced by administration of lipopolysaccharide (LPS) in rats.

METHODS

IN model of rats were induced by lipopolysaccharide. HO activity was determined by the amount of bilirubin formation which was measured with a double-beam spectrophotometer, and HbCO formation was measured by CO-oximeter.

RESULTS

The results showed that LPS (10 mg/kg) caused IN in up to 40% of the rats at 6 h after each treatment of LPS. The incidence of IN were significantly increased by 50% (P < 0.05) and by 83.2% (P < 0.01) in HO substrate (heme-L-lysinate)-treated rats and in exogenous CO-treated rats, respectively; but it was sigificantly decreased by 41.8% (P < 0.05) after administration of ZnDPBG, an inhibitor of heme oxygense (HO) activity. Furthermore, LPS increased HO activity, HbCO formation cGMP content within colic smooth muscle and the plasma level of cGMP, and these parameters were significantly elevated by 62.6% (P < 0.01), 40.0% (P < 0.01), 49.3% (P < 0.05) and 38.9% (P<0.05), respectively, compared with LPS-non-IN rats.

CONCLUSION

It is suggested that endogenous HO/CO system plays an important role in the process of IN induced by LPS, and inhibition of HO activity may decrease the formation of IN.

目的

研究内源性血红素加氧酶(HO)/一氧化碳(CO)系统在调节脂多糖(LPS)诱导的大鼠肠套叠(IN)过程中的作用。

方法

用脂多糖诱导大鼠IN模型。通过用双光束分光光度计测量胆红素形成量来测定HO活性,并用CO血氧计测量HbCO形成量。

结果

结果显示,每次给予LPS后6小时,LPS(10mg/kg)可使高达40%的大鼠发生IN。HO底物(血红素-L-赖氨酸盐)处理的大鼠和外源性CO处理的大鼠中IN的发生率分别显著增加50%(P<0.05)和83.2%(P<0.01);但给予血红素加氧酶(HO)活性抑制剂ZnDPBG后,IN发生率显著降低41.8%(P<0.05)。此外,LPS增加了绞痛平滑肌内的HO活性、HbCO形成、cGMP含量以及血浆cGMP水平,与未发生IN的LPS处理大鼠相比,这些参数分别显著升高62.6%(P<0.01)、40.0%(P<0.01)、49.3%(P<0.05)和38.9%(P<0.05)。

结论

提示内源性HO/CO系统在LPS诱导的IN过程中起重要作用,抑制HO活性可能减少IN的形成。
