To investigate the abnormalities of the relative factors of pRb-p16 cell regulatory pathway in pancreatic adenocarcinoma.
Five strains of human pancreatic adenocarcinoma cell lines were analysed by PCR and single-strand conformation polymorphism (SSCP) for homozygous deletion and mutation of the p16 gene, respectively. The gene amplification of cyclin D1 was detected by Southern blot analysis and the protein expression of pRb gene was examined by Western blot analysis.
Homozygous deletion of exon 1 were found in two cell lines and no mutation was found within exon 1 and exon 2 in five cell lines. Cyclin D1 gene amplification was observed in one cell line and all of the five cell lines showed hyper-phosphorylated pRb protein.
These findings have demonstrated that there are abnormalities of pRb-p16 regulatory pathway in pancreatic adenocarcinoma cell lines, revealing the alteration of the p16 and cyclin D1 genes.

分别采用聚合酶链反应(PCR)和单链构象多态性分析(SSCP)对5株人胰腺腺癌细胞系进行分析,以检测p16基因的纯合缺失和突变情况。通过Southern印迹分析检测细胞周期蛋白D1(cyclin D1)的基因扩增情况,并用Western印迹分析检测pRb基因的蛋白表达情况。
在2个细胞系中发现第1外显子的纯合缺失,在5个细胞系的第1和第2外显子中均未发现突变。在1个细胞系中观察到细胞周期蛋白D1基因扩增,且所有5个细胞系均显示pRb蛋白高度磷酸化。
这些研究结果表明胰腺腺癌细胞系中存在pRb-p16调控通路异常,揭示了p16和细胞周期蛋白D1基因的改变。
