To investigate the effects of fluoride on activities of tartrate-resistant acid phosphate (TRAP) and matrix metalloproteinase-9 (MMP-9) in rat osteoclasts cultured in vitro.
Osteoclast was isolated mechanically from long bones of neonatal rats and cultured in vitro. Histochemical stain was applied to detect the effects of fluoride on activities of TRAP and in-situ hybridization was used to study the expression of MMP-9 mRNA in rat osteoclasts in vitro.
Number of TRAP positive cells was 154.2, 160.0, 170.6, 179.0 and 180.0 per cm(2), respectively for the rats with varied doses of fluoride, in a dose-response pattern but without statistical significance. The expression of MMP-9 mRNA increased with elevating dose of fluoride, especially in the rats with 1.00, 2.00 and 4.00 mg/L of fluoride, to 94.50, 94.64 and 104.97, respectively, significantly different from those in control group.
Fluoride can enhance the MMP-9 mRNA expression in cultured osteoclasts of rats.
研究氟对体外培养的大鼠破骨细胞中抗酒石酸酸性磷酸酶(TRAP)和基质金属蛋白酶-9(MMP-9)活性的影响。
从新生大鼠长骨中机械分离破骨细胞并进行体外培养。采用组织化学染色检测氟对TRAP活性的影响,运用原位杂交技术研究氟对体外培养的大鼠破骨细胞中MMP-9 mRNA表达的影响。
不同剂量氟处理的大鼠,每平方厘米TRAP阳性细胞数分别为154.2、160.0、170.6、179.0和180.0,呈剂量反应关系,但无统计学意义。MMP-9 mRNA表达随氟剂量增加而升高,尤其是氟浓度为1.00、2.00和4.00 mg/L的大鼠,分别升至94.50、94.64和104.97,与对照组相比有显著差异。
氟可增强体外培养的大鼠破骨细胞中MMP-9 mRNA的表达。
