Abstract

OBJECTIVE

To investigate the effects of melatonin, an endogenous hormone, on acetylcholine and KCl-induced contractions of isolated guinea-pig detrusor muscle.

MATERIALS AND METHODS

Detrusor smooth muscle strips isolated from guinea-pig bladders were placed in an organ bath containing physiological saline at 37 degrees C and pH 7.4, constantly bubbled with 95% oxygen and 5% CO2. The effects of cumulatively applied melatonin on the acetylcholine- and KCl-induced contractions of isolated bladder strips were examined using isometric contraction measurements.

RESULTS

Melatonin (100 and 300 micromol/L) significantly inhibited the peak amplitude of both acetylcholine (10 micromol/L) and KCl (30 mmol/L)-induced contraction of the isolated bladder strips (P < 0.05). Similarly, melatonin caused a significant reduction in the contractile frequency induced by KCl (eight strips) in a concentration-dependent manner, while having no significant effect on the frequency of contractile response to acetylcholine, even at the highest concentration (300 micromol/L) used (P = 0.58, 14 strips).

CONCLUSIONS

These results suggest that melatonin inhibits acetylcholine- and KCl-induced contractions in isolated bladder strips from guinea pigs. The endogenous nature of melatonin, with its low side-effect profile, makes it a potentially useful agent to be considered in the medical management of the overactive bladder.

材料与方法

从豚鼠膀胱分离出的逼尿肌平滑肌条置于含37℃生理盐水、pH 7.4的器官浴中,持续用95%氧气和5%二氧化碳鼓泡。使用等长收缩测量法检测累积应用褪黑素对离体膀胱条乙酰胆碱和氯化钾诱导收缩的影响。

结果

褪黑素(100和300微摩尔/升)显著抑制乙酰胆碱(10微摩尔/升)和氯化钾(30毫摩尔/升)诱导的离体膀胱条收缩的峰值幅度(P < 0.05)。同样,褪黑素以浓度依赖性方式显著降低氯化钾(八条平滑肌条)诱导的收缩频率,而即使在所用最高浓度(300微摩尔/升)下,对乙酰胆碱诱导的收缩反应频率也无显著影响(P = 0.58,十四条平滑肌条)。
