To explore the efficiency of liposome-mediated gene transfer into bone marrow cells and the stability of its expression.
Two marker genes (Neo and Lac Z) mediated by liposome were co-transferred into bone marrow cells. Following G418 screening,the positive cells were enriched and expanded in vitro.
The liposome-mediated gene transfer rate in bone marrow cells was 13.33 +/- 2.68%, and after G418 screening, it was 46.06 +/- 3.47%. The blue colouring rates with X-gal staining of these cells were not significantly different before and after expansion.
Liposome-mediated gene transfer in bone marrow cells is highly efficient and the expression of the transferred gene is stable.
探讨脂质体介导的基因导入骨髓细胞的效率及其表达的稳定性。
将脂质体介导的两种标记基因(Neo和Lac Z)共导入骨髓细胞。经G418筛选后,富集阳性细胞并在体外扩增。
脂质体介导的骨髓细胞基因转染率为13.33±2.68%,经G418筛选后为46.06±3.47%。这些细胞经X-gal染色后的蓝色着色率在扩增前后无显著差异。
脂质体介导的基因导入骨髓细胞效率高,且导入基因的表达稳定。
